Here we used time-correlated single-photon countingfluorescence lifetime imaging microscopy (FLIM) to detect changes in donorlifetime following the photoactivation of PA-GFP. The time-domain FLIMmeasurements from cells expressing CFP-C/EBP[alpha] alone indicated anaverage fluorescence lifetime of about 2.14 ns, which was unaffected when wephotoactivated these cells (Table 1). For cells that coexpressedCFP-C/EBP[alpha] and PA-GFP-HP1[alpha], upon photoactivation of PA-GFP themean CFP lifetime distribution shifted to shorter times (1.82 ns; Table 1 andSupplementary Fig. 4 online). Additionally, the shift to shorter donorlifetimes was more pronounced in cells that coexpressed CFP-C/EBP BZIP andPA-GFP-C/EBP BZIP (1.60 ns; Table 1 and Supplementary Fig. 4).
